Background
OX40L,amemberoftheTNFsuperfamilyofstructurallyrelatedproteins,existsprimarilyasatypeIImembranebound,non-covalentlylinkedhomotrimericprotein.Itisexpressedonantigenpresentingcells(APCs),suchasdendriticcellsandactivatedB-cells,andalsoonvariousothercellssuchasvascularendothelialcells,mastcells,andnaturalkillercells.OX40LsignalsspecificallythroughtheOX40receptor,whichisexpressedpredominantlyonCD4+TcellsbutalsooncertainactivatedCD8+Tcells.OX40/OX40Lfunctionsasacostimulatorysignal,whichisrequiredforaproductiveinteractionbetweenantigenpresentingcellsandtheirtargetT-cells.Itenhancescellproliferationandsurvival,andincreasesexpressionofRANTES,IL-2,IL-3,andIFNγ.OX40/OX40Lsignalingplaysanimportantroleinimmuno-regulatorycommunication,enablingtheimmunesystemtodistinguishbetween“friendvs.foe”duringactivation;amechanismtypicallytermedimmuno-tolerance.RecombinantOX40Lisaglycosylated133aminoacidproteincorrespondingtotheextracellularTNFhomologousdomainofthefulllengthtransmembraneprotein.Itmigrateswithanapparentmolecularmassof15.5–25.0kDaonSDS-PAGE.
